What Are Interleukins?

Interleukins (ILs) are a large family of cytokines — small signaling proteins primarily secreted by leukocytes — that mediate communication between immune cells. First discovered in the 1970s, interleukins regulate immune cell growth, differentiation, activation, and migration, playing central roles in both innate and adaptive immunity.

Each interleukin binds to specific receptors on target cells to trigger intracellular signaling cascades such as JAK-STAT, MAPK, and NF-κB pathways. Because of their central role in immune regulation, recombinant interleukins are essential research reagents for studying immune cell biology, developing immunotherapies, and screening drug candidates.

Types of Interleukins & Their Functions

IL-1 Family

The IL-1 family (IL-1α, IL-1β, IL-1RA, IL-18, IL-33, IL-36, IL-37, IL-38) is a central mediator of inflammation and host defense. IL-1β is a key therapeutic target in autoinflammatory diseases; IL-33 drives type 2 immune responses in allergy and asthma research.

IL-2 Family (Common γ-Chain Cytokines)

IL-2, IL-4, IL-7, IL-9, IL-15, and IL-21 share the common gamma chain (γc/CD132) receptor subunit. IL-2 is critical for regulatory T cell (Treg) maintenance and is widely used in T cell culture protocols. IL-15 supports NK cell expansion — both are essential reagents in CAR-T and NK cell therapy development.

IL-6 Family

IL-6, IL-11, IL-27, IL-31, LIF, OSM, and CNTF signal through the gp130 receptor. IL-6 is a pleiotropic cytokine involved in inflammation, acute phase response, and B cell differentiation — a major drug target validated by anti-IL-6R therapies (tocilizumab).

IL-10 Family

IL-10, IL-19, IL-20, IL-22, IL-24, and IL-26 regulate immune tolerance and tissue homeostasis. IL-10 is the prototypic anti-inflammatory cytokine, suppressing pro-inflammatory responses from macrophages and dendritic cells.

IL-12 Family

IL-12, IL-23, IL-27, and IL-35 are heterodimeric cytokines that bridge innate and adaptive immunity. IL-12 promotes Th1 differentiation and is central to anti-tumor immunity research; IL-23 is a validated target in psoriasis and inflammatory bowel disease.

IL-17 Family

IL-17A, IL-17B, IL-17C, IL-17D, IL-17E (IL-25), and IL-17F mediate neutrophil recruitment and mucosal immunity. IL-17A is the target of secukinumab and ixekizumab — blockbuster therapies for psoriasis and ankylosing spondylitis.

Applications of Interleukins in Research & Medicine

  • Cancer immunotherapy: IL-2 and IL-15 for T cell and NK cell expansion in adoptive cell therapy; IL-12 as an investigational anti-tumor cytokine
  • Autoimmune disease research: IL-17, IL-23, and IL-6 as drug targets and assay standards in psoriasis, RA, and IBD studies
  • Infectious disease & vaccine development: Cytokine profiling and adjuvant research using recombinant IL standards
  • Stem cell & regenerative medicine: IL-6, LIF, and IL-3 for cell differentiation and expansion protocols
  • Immunoassay development: High-purity IL proteins as coating antigens, calibration standards, and positive controls for ELISA
