What Are Fc Receptors?
Fc receptors bind the constant (Fc) region of antibodies, bridging humoral and cellular immunity. They determine antibody effector functions — antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and complement activation — making them central to therapeutic antibody development.
Key Fc Receptors
Fcγ Receptors (FcγRI/II/III)
Bind IgG with varying affinities. FcγRIIIa (CD16a) engagement is the primary driver of NK cell-mediated ADCC; its V158F polymorphism affects clinical response to rituximab and trastuzumab. Recombinant FcγRs are essential for ADCC reporter assays and Fc engineering studies.
FcRn
The neonatal Fc receptor recycles IgG, determining antibody half-life. FcRn binding assays at pH 6.0 vs 7.4 are standard in antibody engineering — mutations (YTE, LS) that enhance FcRn binding extend therapeutic antibody half-life.
Fcε Receptors
Bind IgE and mediate allergic responses; targets of omalizumab-based strategies and mast cell research.
