RuvA-RuvB increases the rate of strand exchange (branch migration), dissociates the RecA filament and allows RuvC to cleave in both orientations at the cruciform junction. HJ-RuvA-RuvB-RuvC complexes resolve Holliday junctions and also undergo branch migration, providing evidence for a coupled branch migration/HJ resolution reaction.

Homodimer which binds Holliday junction (HJ) DNA. The HJ becomes 2-fold symmetrical on binding to RuvC with unstacked arms; it has a different conformation from HJ DNA in complex with RuvA. In the full resolvosome a probable DNA-RuvA(4)-RuvB(12)-RuvC(2) complex forms which resolves the HJ (By similarity). A mix of monomer and homodimer in solution (PubMed:8106500). Homodimer which binds DNA; there is probably a monomer-dimer equilibrium during DNA-binding (PubMed:10471285, PubMed:1661673, PubMed:7638215, PubMed:7923356, PubMed:9135161). Can form complexes with RuvA and Holliday junction (HJ) DNA, in which the resolvase activity is inhibited (PubMed:9000618). Forms a complex with RuvB without DNA; RuvA-RuvC complexes were not detected in the absence of DNA (PubMed:9160752). In the presence of HJ DNA a DNA-RuvA-RuvB-RuvC complex forms with 2 rings of RuvB that resolves upon addition of ATP. Upon resolution the protein complex dissociates from DNA (PubMed:10421637, PubMed:9637927)
