Function

Member of the two-component regulatory system PhoP/PhoQ involved in virulence, adaptation to low Mg(2+) environments and the control of acid resistance genes. Also involved in adaptation to hyperosmotic environments, in a Mg(2+)-independent manner. In low periplasmic Mg(2+), PhoQ functions as a membrane-associated protein kinase that undergoes autophosphorylation and subsequently transfers the phosphate to PhoP, resulting in the expression of PhoP-activated genes (PAG) and repression of PhoP-repressed genes (PRG).

Biological Context

Subcellular Location: Cell inner membrane (Multi-pass membrane protein)

Product Specifications

Recombinant Escherichia coli O6:H1 Sensor protein PhoQ (phoQ) Protein is a recombinant protein from Escherichia coli O6:H1 (strain CFT073 / ATCC 700928 / UPEC), expressed in in vitro E.coli expression system, covering amino acids 1-486aa, with N-terminal 10xHis-tagged tag, molecular weight 56.8kDa, purity Greater than 85% as determined by SDS-PAGE.. Suitable for ELISA and Western Blot applications.

Recommended applications depend on the target, product format, and product-specific validation data. Refer to the product datasheet or Certificate of Analysis (CoA), or contact BioCrest Sci for application guidance.

Protein Biology

Function

Member of the two-component regulatory system PhoP/PhoQ involved in virulence, adaptation to low Mg(2+) environments and the control of acid resistance genes (By similarity). Also involved in adaptation to hyperosmotic environments, in a Mg(2+)-independent manner (By similarity). In low periplasmic Mg(2+), PhoQ functions as a membrane-associated protein kinase that undergoes autophosphorylation and subsequently transfers the phosphate to PhoP, resulting in the expression of PhoP-activated genes (PAG) and repression of PhoP-repressed genes (PRG) (By similarity). In high periplasmic Mg(2+), acts as a protein phosphatase that dephosphorylates phospho-PhoP, which results in the repression of PG and may lead to expression of some PRG (By similarity)

Subcellular Location

Cell inner membrane (Multi-pass membrane protein)

Subunit

Homodimer; probably dimerizes via the cytoplasmic domain. Probably interacts with MgrB in the periplasm, altering its activity and that of downstream effector PhoP. Interacts with two-component system connector protein SafA (via C-terminus)

Gene: phoQ  |  Organism: Escherichia coli O6:H1 (strain CFT073 / ATCC 700928 / UPEC)  |  Synonyms: Sensor histidine protein kinase/phosphatase PhoQ
