Interleukins have moved from basic immunology to the center of cancer therapy. IL-18 and IL-21: Emerging Players

IL-18 synergizes with IL-12 to drive IFN-γ production; an engineered “decoy-resistant” IL-18 (DR-18) shows remarkable anti-tumor activity in preclinical models. IL-21 supports CD8+ T cell and NK cell function and has been explored in combination with checkpoint inhibitors.
