Function

Single-stranded DNA-binding protein that participates in viral DNA replication, formation of concatemers, recombination and repair of double-stranded breaks. Coats the lagging-strand ssDNA as the replication fork advances and stimulates the activities of viral DNA polymerase and primase/helicase. Coordinates simultaneous synthesis of leading- and lagging-strands.

Product Specifications

Recombinant Escherichia phage T7 Single-stranded DNA-binding protein (2.5) is a recombinant protein from Escherichia phage T7 (Bacteriophage T7), expressed in Yeast, covering amino acids 1-232aa, with Tag-Free tag, molecular weight 27.7kDa, purity Greater than 90% as determined by SDS-PAGE.. Suitable for ELISA and Western Blot applications.

SDS-PAGE: Single band at expected molecular weight confirming purity.

ELISA: Suitable as coating antigen or detection standard.

Western Blot: Compatible with standard Western Blot protocols.

Protein Interaction: Validated for SPR (Surface Plasmon Resonance) and BLI (Bio-Layer Interferometry) studies.

Shipping: Shipped at ambient temperature. Lyophilized protein is stable during transit.

Storage: Store lyophilized protein at -20°C to -80°C. Reconstituted protein should be aliquoted and stored at -80°C. Avoid repeated freeze-thaw cycles.

Protein Biology

Function

Single-stranded DNA-binding protein that participates in viral DNA replication, formation of concatemers, recombination and repair of double-stranded breaks (PubMed:11222583, PubMed:16807232, PubMed:8617248, PubMed:9079662). Coats the lagging-strand ssDNA as the replication fork advances and stimulates the activities of viral DNA polymerase and primase/helicase (PubMed:8617248). Coordinates simultaneous synthesis of leading- and lagging-strands (PubMed:9651583). Together with DNA primase/helicase, promotes pairing of two homologous DNA molecules containing complementary single-stranded regions and mediates homologous DNA strand exchange (PubMed:8617248). Also promotes the formation of joint molecules (PubMed:8617248, PubMed:9079662). Disrupts loops, hairpins and other secondary structures present on ssDNA to reduce and eliminate pausing of viral DNA polymerase at specific sites during elongation (PubMed:1634538)

Subunit

Homodimer (PubMed:11481454, PubMed:1634538). Interacts (via C-terminus) with the viral DNA polymerase (PubMed:12766155, PubMed:15795374, PubMed:1634539, PubMed:8106511). Interacts with the viral helicase/primase (PubMed:1634539). Part of the replicase complex that includes the DNA polymerase, host thioredoxin, the primase/helicase and the single-stranded DNA binding protein (PubMed:22977246)
