Function

Class II viral fusion protein. Fusion activity is inactive as long as E1 is bound to E2 in mature virion. After virus attachment to target cell via host VLDLR or LRP8/APOER2 and endocytosis, acidification of the endosome induces dissociation of E1/E2 heterodimer and concomitant trimerization of the E1 subunits.

Biological Context

Subcellular Location: Virion membrane (Single-pass type I membrane protein); Host cell membrane (Single-pass type I membrane protein)

Product Specifications

Recombinant Sindbis virus Structural polyprotein, partial Protein is a recombinant protein from Sindbis virus (SINV), expressed in in vitro E.coli expression system, covering amino acids 807-1245aa, with N-terminal 10xHis-SUMO-tagged tag, molecular weight 65.9kDa, purity Greater than 85% as determined by SDS-PAGE.. Suitable for ELISA and Western Blot applications.

SDS-PAGE: Single band at expected molecular weight confirming purity.

ELISA: Suitable as coating antigen or detection standard.

Western Blot: Compatible with standard Western Blot protocols.

Protein Interaction: Validated for SPR (Surface Plasmon Resonance) and BLI (Bio-Layer Interferometry) studies.

Shipping: Shipped at ambient temperature. Lyophilized protein is stable during transit.

Storage: Store lyophilized protein at -20°C to -80°C. Reconstituted protein should be aliquoted and stored at -80°C. Avoid repeated freeze-thaw cycles.

Shelf Life: 12 months from date of receipt when stored as recommended.

Protein Biology

Function

Class II viral fusion protein. Fusion activity is inactive as long as E1 is bound to E2 in mature virion (PubMed:12573591, PubMed:21124457). After virus attachment to target cell via host VLDLR or LRP8/APOER2 and endocytosis, acidification of the endosome induces dissociation of E1/E2 heterodimer and concomitant trimerization of the E1 subunits (PubMed:21124457, PubMed:34929721). This E1 trimer is fusion active, and promotes release of viral nucleocapsid in cytoplasm after endosome and viral membrane fusion (PubMed:21124457). Efficient fusion requires the presence of cholesterol and sphingolipid in the target membrane (PubMed:10482600)

Subcellular Location

Virion membrane (Single-pass type I membrane protein); Host cell membrane (Single-pass type I membrane protein)

Subunit

Oligomer (By similarity). Interacts with spike glycoprotein E1 (PubMed:8892914). Interacts with spike glycoprotein E2 (PubMed:8892914)

Organism: Sindbis virus  |  Synonyms: p130
