Function

Viral CC chemokine that binds to a broad spectrum of viral and human chemokine receptors and, depending on the receptor, acts either as an antagonist or an agonist. Capable of binding to chemokine receptors of the 4 families (CCR, CXCR, XCR, and CX3CR). Acts as an antagonist toward CCR1, CCR2, CCR5, and CCR10, competing with the endogenous chemokine binding and signaling.

Product Specifications

Recombinant Human herpesvirus 8 type P Viral macrophage inflammatory protein 2 (VMI2), partial (Active) is a recombinant protein from Human herpesvirus 8 type P (isolate GK18) (HHV-8) (Kaposi's sarcoma-associated herpesvirus), expressed in E.Coli, covering amino acids 24-93aa, with Tag-Free tag, molecular weight 8.0 kDa, purity >97% as determined by SDS-PAGE.. Suitable for ELISA and Western Blot applications.

SDS-PAGE: Single band at expected molecular weight confirming purity.

ELISA: Suitable as coating antigen or detection standard.

Western Blot: Compatible with standard Western Blot protocols.

Protein Interaction: Validated for SPR (Surface Plasmon Resonance) and BLI (Bio-Layer Interferometry) studies.

Shipping: Shipped at ambient temperature. Lyophilized protein is stable during transit.

Storage: Store lyophilized protein at -20°C to -80°C. Reconstituted protein should be aliquoted and stored at -80°C. Avoid repeated freeze-thaw cycles.

Protein Biology

Function

Viral CC chemokine that binds to a broad spectrum of viral and human chemokine receptors and, depending on the receptor, acts either as an antagonist or an agonist. Capable of binding to chemokine receptors of the 4 families (CCR, CXCR, XCR, and CX3CR). Acts as an antagonist toward CCR1, CCR2, CCR5, and CCR10, competing with the endogenous chemokine binding and signaling (PubMed:9287217). On the other hand, acts as an agonist of CCR3 and CCR8, triggering G protein-mediated intracellular calcium release and activation of the ERK and PI3K/AKT signaling pathways, or chemotaxis (PubMed:9323208, PubMed:9834206, PubMed:11500830). Thereby, participates in the skewing of the host immune system away from the Th1 cytotoxic response, undesirable for the virus-infected cells, toward a Th2 response, which is less effective against intracellular pathogens (PubMed:9834206, PubMed:11500830). CXCR7 acts as a scavenger receptor for vCCL2, regulating its availability and activity toward other host receptors (PubMed:27238288)

Subunit

Monomer. Interacts with human chemokine receptor CXCR4 (PubMed:25612609). Interacts with host CCR3 (PubMed:9323208, PubMed:11500830). Interacts with host CCR8 (PubMed:9834206). Interacts with host CCR1, CCR2 and CCR5 (PubMed:9287217). Interacts with ACKR3/CXCR7; this interaction regulates vCCL2 availability and activity toward human receptors (PubMed:27238288)
